    Soil and Water Conservation and Integrated Management in Watershed Ecosystem of the Loess Plateau, China

    The Loess Plateau is well known for severe soil erosion and ecosystem degradation. The key issues in this region are the food security (grain), ecological security and a sustainable economy. This paper reviews a range of studies on ecological restoration, and management and the benefit at watershed scale on the Loess Plateau. This includes several successful cases and failures in soil conservation. The paper presents practical soil conservation measures and related benefit analysis, and discusses some effective methods adopted in soil erosion control, research directions and future perspectives for the Loess Plateau

    DESIGN AND OPTIMIZATION OF THE VARIABLE-DENSITY LATTICE STRUCTURE BASED ON LOAD PATHS

    Lattice structure is more and more widely used in engineering by replacing solid structure. But its mechanical performances are constrained by the external shape if the unit cells are directly filled in the design domain, and the traditional topology optimization methods are difficult to give the explicitly mechanical guidance for the distribution of internal unit cells. In this paper, a novel design and optimization method of variable-density lattice structure is proposed in order to simultaneously optimize the external shape and the internal unit cells. First of all, the envelope model of any given structure should be established, and the load paths need to be visualized by the theory of load path. Then, the design criteria of external shape are established based on the principle of smoother load paths in the structure. An index of load flow capacity is defined to indicate the load paths density and to control the density distribution of unit cells, and a detailed optimization strategy is given. Finally, three examples of a cantilever plate, an L-shaped bracket and a classical three-point bending beam are used to verify the method. The results show that the models designed by the proposed method have better mechanical performances, lower material usage and less printing time

    SOX7 is involved in polyphyllin D-induced G0/G1 cell cycle arrest through down-regulation of cyclin D1

    The incidence of mortality of prostate cancer (PCa) has been an uptrend in recent years. Our previous study showed that the sex-determining region Y-box 7 (SOX7) was low-expressed and served as a tumor suppressor in PCa cells. Here, we describe the effects of polyphyllin D (PD) on proliferation and cell cycle modifications of PCa cells, and whether SOX7 participates in this process. PC-3 cells were cultured in complete medium containing PD for 12, 24, and 48 h. MTT assay was used to investigate the cytotoxic effects of PD. Cell cycle progression was analyzed using propidium iodide (PI) staining, and protein levels were assayed by Western blot analysis. Our results showed low expression of SOX7 in PCa tissues/cells compared to their non-tumorous counterparts/RWPE-1 cells. Moreover, PD inhibited the proliferation of PC-3 cells in a dose- and time-dependent manner. PD induced G0/G1 cell cycle arrest, while co-treatment with short interfering RNA targeting SOX7 (siSOX7) had reversed this effect. PD downregulated SOX7, cyclin D1, cyclin-dependent kinase 4 (CDK4), and cyclin-dependent kinase6 (CDK6) expressions in a dose-dependent manner, whereas co-treatment of siSOX7 and PD rescued the PD-inhibited cyclin D1 expression. However, no obvious changes were observed in CDK4 or CDK6 expression. These results indicate that SOX7 is involved in PD-induced PC-3 cell cycle arrest through down-regulation of cyclin D1

    CoRide: Joint Order Dispatching and Fleet Management for Multi-Scale Ride-Hailing Platforms

    How to optimally dispatch orders to vehicles and how to tradeoff between immediate and future returns are fundamental questions for a typical ride-hailing platform. We model ride-hailing as a large-scale parallel ranking problem and study the joint decision-making task of order dispatching and fleet management in online ride-hailing platforms. This task brings unique challenges in the following four aspects. First, to facilitate a huge number of vehicles to act and learn efficiently and robustly, we treat each region cell as an agent and build a multi-agent reinforcement learning framework. Second, to coordinate the agents from different regions to achieve long-term benefits, we leverage the geographical hierarchy of the region grids to perform hierarchical reinforcement learning. Third, to deal with the heterogeneous and variant action space for joint order dispatching and fleet management, we design the action as the ranking weight vector to rank and select the specific order or the fleet management destination in a unified formulation. Fourth, to achieve the multi-scale ride-hailing platform, we conduct the decision-making process in a hierarchical way where a multi-head attention mechanism is utilized to incorporate the impacts of neighbor agents and capture the key agent in each scale. The whole novel framework is named as CoRide. Extensive experiments based on multiple cities real-world data as well as analytic synthetic data demonstrate that CoRide provides superior performance in terms of platform revenue and user experience in the task of city-wide hybrid order dispatching and fleet management over strong baselines.Comment: CIKM 201

    Identification of SNPs in MITF associated with beak color of duck

    Introduction: Beak color—a pigment-related trait—is an important feature of duck breeds. Recently, little research has addressed genetic mechanism of the beak colors in poultry, whereas the process and the regulation factors of melanin deposition have been well described.Methods: To investigate the genetic mechanism of beak colors, we conducted an integrated analysis of genomic selection signatures to identify a candidate site associated with beak color. For this, we used black-billed (Yiyang I meat duck synthetic line H1, H2, H3&HF) and yellow-billed ducks (Cherry Valley ducks and white feather Putian black duck). Quantitative real-time PCR and genotyping approaches were used to verify the function of the candidate site.Results: We identified 3,895 windows containing 509 genes. After GO and KEGG enrichment analysis, nine genes were selected. Ultimately, MITF was selected by comparing the genomic differentiation (FST). After loci information selection, 41 extreme significantly different loci were selected, which are all located in intron regions of MITF and are in almost complete linkage disequilibrium. Subsequently, the site ASM874695v1:10:g.17814522T > A in MITF was selected as the marker site. Furthermore, we found that MITF expression is significantly higher in black-beaked ducks than in yellow-beaked ducks of the F2 generation (p < 0.01). After genotyping, most yellow-billed individuals are found with homozygous variant; at the same time, there are no birds with homozygous variant in black-billed populations, while the birds with homozygous and heterozygous variant share the same proportion.Conclusion:MITF plays a very critical role in the melanogenesis and melanin deposition of duck beaks, which can effectively affect the beak color. The MITF site, ASM874695v1:10:g.17814522T > A could be selected as a marker site for the duck beak color phenotype
